Have you also encountered the following problems during the process of frozen slicing:
Blurred staining of the cell nucleus, degeneration of cellular tissue, incomplete fragmentation of slices, curled and wrinkled slices, and the appearance of stripes and folds
Possible reasons:
REASON
1. Insufficient dehydration of gradient ethanol after staining resulted in liquid entering the transparent solution of the slices, leading to incomplete transparency. Observe the cloud like appearance with the naked eye. The tissue structure is blurred under the microscope.
2. The liquid level of the transparent liquid is low and has not reached the height of the slice gradient ethanol, causing the ethanol liquid to be carried into the transparent liquid
resolvent:
METHOD
The sliced tissue was not completely removed by anhydrous ethanol in the clear solution, and was returned to anhydrous ethanol. The clear solution was replaced to ensure the standard liquid level before being re transparent.
