Hematoxylin staining is too light
Possible causes:
① Hematoxylin solution is too aging, low room temperature, insufficient hematoxylin oxidation is not easy to color;
② The differentiation time is too long or the concentration of differentiation liquid is high;
(3) The dyeing time of hematoxylin solution is short;
④ Poor fixation treatment, tissue is not easy to stain;
⑤ the degree of bluing is not enough;
The solution:① Standardize the use of relatively stable commercial hematoxylin, monitor room temperature changes, or replace fresh hematoxylin solution; ② Reduce the differentiation time or dilute the differentiation fluid; ③ Prolong hematoxylin dyeing time; ④ High concentration alcohol fixed time is too long, can not dye color, with 2% iron alum treatment. ⑤ Replace the new blue solution, such as ammonia or use Beant medical special H-E HD constant dyeing set, the blue return effect is stable; If you use tap water to do back blue, you need to test whether the tap water is weakly alkaline.
The eosin stain is too light
Possible causes:
① Eosin dye is used for too long and needs to be replaced;
② Alcohol washing after eosin dye solution is incorrect;
③ eosin dyeing time is too short;
The solution:① Replace fresh eosin dye solution; ② Reduce the time of alcohol washing after eosin step; ③ Increase the dyeing time of eosin solution.
Uneven dyeing
Possible causes:
① Before the dyeing machine enters Eosin, a part of the tissue surface is attached with grease, which is the dewaxing liquid of the dyeing frame when the dyeing machine is running in the dyeing tank (generally, it is easy to attach to the slide in the blue liquid), and the grease attached to the tissue will not be stained when it enters eosin dyeing.
The quality problem of fixed liquid: the commonly used simple fixed liquid formaldehyde solution is itself a reducing agent, which is extremely volatile and has a strong irritating odor. The cells fixed with formaldehyde solution had better nuclear staining but worse cytoplasm staining.
③ Due to the repeated use of the slicer, there are some customer errors: subjective errors caused by human operation, so that the slice is not uniform, therefore, the dyeing is not uniform.
④ Incomplete dewaxing agent and gradient ethanol have impurities, low room temperature, and long use time, resulting in unclean dewaxing.
